Meer onderzoek naar de SEAL gentherapie

Wat lossen we met dit project op?

Voor mensen met LAMA2-RD bestaat nog geen behandeling die de oorzaak van de ziekte aanpakt. Hoewel wereldwijd verschillende therapieën worden onderzocht, zijn veel daarvan alleen geschikt voor een beperkte groep patiënten met een specifieke genetische mutatie. De SEAL-gentherapie is juist ontwikkeld om in de toekomst geschikt te zijn voor vrijwel alle mensen met LAMA2-RD. Met dit project willen we voorkomen dat dit veelbelovende onderzoek stilvalt door een gebrek aan financiering.

Wat houdt dit project precies in en wie voert het uit?

Professor Markus Rüegg en zijn team aan de Universiteit van Basel werken al meer dan 25 jaar aan de ontwikkeling van de SEAL-gentherapie. Deze innovatieve behandeling maakt gebruik van twee AAV-vectoren die samen het ontbrekende LAMA2-eiwit kunnen herstellen. In dieronderzoek zijn zeer veelbelovende resultaten behaald, met een sterk herstel van de spier- en zenuwfunctie. De komende onderzoeksfase is gericht op het verder optimaliseren van de therapie en het zetten van de noodzakelijke stappen richting klinische studies bij patiënten.

Voor wie is dit project bedoeld?

Dit onderzoek is bedoeld voor iedereen met LAMA2-RD. Omdat de therapie niet afhankelijk is van een specifieke mutatie, heeft zij de potentie om in de toekomst geschikt te zijn voor vrijwel alle patiënten, ongeacht hun genetische afwijking.

Waarom vindt Voor Sara dit een belangrijk project?

Stichting Voor Sara gelooft dat de grootste kans op een behandeling ontstaat door veelbelovende onderzoeken actief te ondersteunen. Daarom investeerden wij eerder al ruim €45.000 in dit project. Die bijdrage maakte belangrijke vervolgstappen mogelijk. Inmiddels werken we samen met internationale patiëntenorganisaties, onderzoekers en potentiële financiers om ook de volgende onderzoeksfase mogelijk te maken. Ons doel is om opnieuw financieel bij te dragen en zo de ontwikkeling van deze therapie te versnellen.

Waar staan we nu met dit project?

De eerste onderzoeksfase heeft veelbelovende resultaten opgeleverd. Nu is het moment aangebroken om de therapie verder door te ontwikkelen en voor te bereiden op de volgende stap richting klinische studies. Hiervoor is aanvullende financiering nodig. Samen met internationale partners werken we aan een plan om deze financiering rond te krijgen en het onderzoek voort te zetten.

Wat kost dit project?

De exacte kosten van de volgende onderzoeksfase worden momenteel uitgewerkt door het onderzoeksteam. Het gaat om een omvangrijk internationaal traject waarvoor enkele miljoenen euro’s nodig zullen zijn. Stichting Voor Sara wil hierin opnieuw een substantiële bijdrage leveren en daarnaast andere fondsen en financiers aan dit project verbinden.

Advancing Research into the SEAL Gene Therapy

What problem does this project solve?

There is currently no treatment that addresses the underlying cause of LAMA2-RD. Although several therapies are being developed around the world, many of them are only suitable for a small group of patients with specific genetic mutations. The SEAL gene therapy has been designed with the goal of benefiting nearly everyone living with LAMA2-RD, regardless of their mutation. This project aims to ensure that this promising research can continue and is not delayed due to a lack of funding.

What does this project involve and who is carrying it out?

Professor Markus Rüegg and his team at the University of Basel have been developing the SEAL gene therapy for more than 25 years. This innovative approach uses two AAV vectors that work together to restore the function of the missing LAMA2 protein. In preclinical studies, the therapy has shown highly promising results, with significant improvements in muscle and nerve function. The next phase of the project focuses on further optimizing the therapy and completing the critical steps needed before clinical trials in patients can begin.

Who is this project for?

This research is intended for everyone living with LAMA2-RD. Because the therapy is not mutation-specific, it has the potential to become a treatment for nearly all people affected by the disease, regardless of their genetic variant.

Why does Voor Sara consider this an important project?

Stichting Voor Sara believes that supporting the most promising research offers the best chance of finding an effective treatment for LAMA2-RD. We have already invested more than €45,000 in this project, enabling the research team to make important progress. Today, we are working together with international patient organizations, researchers, and potential funding partners to secure the next phase of development. Our goal is to make a second financial contribution and help accelerate this promising therapy toward patients.

Where does the project stand today?

The first phase of the research has produced very encouraging results. The project has now reached a critical stage in which the therapy must be further developed and prepared for the next steps toward clinical trials. Additional funding is needed to continue this work. Together with international partners, we are developing a funding strategy to ensure the research can move forward.

How much does this project cost?

The budget for the next phase is currently being finalized by the research team. This is a large international development program that will require several million euros. Stichting Voor Sara aims to make another significant financial contribution while also bringing together additional foundations, donors, and funding partners to support this important work.
